Quantification of Venlafaxine in Tablet Dosage Form New chiral normal phase UFLC method for determination of venlafaxine hydrochloride enantiomers in pharmaceutical formulations ABSTRACT Aim: A simple, specific, precise, sensitive and rapid normal phase-UFLC method was developed for determination of venlafaxine hydrochloride enantiomers in pharmaceutical formulation. Methodology: The method was developed on a Lux amylase 2 column (150 x 4.0 mm I.D., particle size 5 ÃŽ ¼); the mobile phase was n-hexane and ethanol (97:3 v/v); in 0.1%diethyamine using UV detector was fixed at 254 nm with a flow rate was 1 mL/min. Results: The retention time (tR) of R- venlafaxine hydrochloride and S- venlafaxine hydrochloride were found to be 4.5 ±0.2 min and 5.3 ±0.3 min, respectively. The linearity over the concentration range of 5-30  µg mL-1 for venlafaxine. The intra-day and inter-day coefficient of variation of the assay method were found to be 0.293 to 1.760 and 0.319 to 0.210 respectively, with high accuracy and precision results. The proposed NP-UFLC method is suitable for analysis of venlafaxine hydrochloride enantiomers in pharmaceutical dosage forms. Conclusion: The validated NP-UFLC method was developed for the quantification of venlafaxine in tablet dosage form. Keywords: R-venlafaxine hydrochloride, S-venlafaxine hydrochloride, enantiomers, NP-UFLC, Validation INTRODUCTION Venlafaxine is a second-generation antidepressant drug marketed as a racemic mixture (Figure 1). The R-enantiomer exhibits dual presynaptic inhibition of serotonin and noradrenaline uptake, whereas the S-enantiomer is a serotonine reuptake inhibitor. Thus, the drug is the first and most commonly used serotonin and noradrenaline reuptake inhibitor. Its synthesis and that of several analogues were described many years ago. The synthetic routes are similar and vary according to the nature of the aromatic substituents. However, the final products are racemic mixtures, and they were crystallized as hydrochlorides.[1] Although the disposition of venlafaxine in humans was originally found not to be stereoselective.[2] In view of the near expiration date (June 2008) of the first patent for the racemic compound and of these recent clinical findings, venlafaxine appears to be a good candidate for a chiral switch.[3-4] Figure 1: Molecular structure of Venlafaxine The trend toward single enantiomer drugs is clear and the number of racemic drugs that reach the market as new chemical entities is decreasing.[5] The relevance of chirality in antidepressant drugs was highlighted several years ago and many examples are illustrated in a recent very complete review.[6-7] In the previously cited research on the resolution of venlafaxine, the enantiomers were separated by either of two general approaches. The first is the classical method of diastereoisomeric salt formation and fractional crystallization and the second approach uses analytical enantioselective electro driven methods. In the latter cases, either cyclodextrinsin capillary electrophoresis.[8] There is only one literature report where an HPLC baseline separation of the enantiomers of venlafaxine extracted was achieved using a CSP and normalphase mode.[9] From an analytical point of view, enantioselective chromatography offers the advantages of a method that can be developed on a semiprepara tive or preparative scale for the isolation of single enantiomers, which then become available for pharmaceutical testing strategies and requirements for enantioselective.[10] In the present research work, a simple, sensitive and accurate normal phase UFLC method to separate R and S-enantiomer of venlafaxine in bulk drugs and tablets using Lux amylase 2 column column has been reported for first time. The method was also validated to ensure the compliance in accordance with the ICH guidelines. MATERIALS AND METHODS Chemicals and Reagents: Venlafaxine hydrochloride enantiomers were a gift sample from R N FINE CHEMICALS BANGALURU, India. The solvents like n-hexane and ethanol diethylamine used was of HPLC grade (Merck, India). Commercially available racemic venlafaxine hydrochloride tablets claimed to contain 25mg of drug were procured from local market. Instrumentation: Quantitative NP-UFLC was performed on gradient high pressure liquid chromatography (Shimadzu) auto sampler consisting of a LC–20HT solvent module, SPD– 10A, and an PDA detector with LC software. The column used was LUX amylase 2 chiral column(150 x 4.0 mm ) particle size 5 ÃŽ ¼. UFLC conditions: The composition of the mobile phase was n-hexane and ethanol in the ratio of 97:03 v/v. They were filtered before use through a 0.2 mm membrane filter, degassed in a bath sonicator for 10 min. The mobile phase was pumped from the solvent reservoir to the column at a flow rate of 1mL/min, which yielded a column backpressure of 96 kg/cm2. The run time was set at 20 min and column temperature was ambient. The volume of injection loop was 20 mL. prior to injection of drug solutions, the column was equilibrated for at least 30 min with the mobile phase flowing through the system. The eluents were monitored at 254 nm and data was acquired, stored and analyzed with the LC 10 software. REAGENTS USED Mobile phase n-hexane and ethanol of HPLC grade was taken as mobile phase in the ratio of 97:3 % (v/v). Preparation of standard stock solution Standard stock solution (100  µg mL-1) of Venlafaxine hydrochloride was prepared by weighing exactly 10 mg of drug dissolved in isopropanol and diluted to 100 mL with same solvent. Preparation of calibration curve Aliquots of Venlafaxine hydrochloride ranging from 0.5-3 mL (each mL contains 100  µg mL-1) were pipetted into as a series of 10mL volumetric flasks. The volume was made up to the mark at with isopropanol. Aliquoets of 10 µL was injected (six time) into HPLC. The elution of the drug measured at 254.0 nm. The amount of venlafaxine hydrochloride present in the sample solution was computed from its calibration curve and it was constructed by plotting peak area of chromatogram against the concentration of Venlafaxine hydrochloride. The blank chromatogram and standard drug chromatogram were shown in figure 2 and 3 respectively. Linearity was 5.0-30  µg mL-1 for Venlafaxine hydrochloride was shown in figure 4. Powder equivalent to 125 mg of venlafaxine hydrochloride was weighed and transferred to a standard volumetric flask. The contents were mixed thoroughly and filtered through a 0.45 ÃŽ ¼m membrane filter. 10 ÃŽ ¼L of the sample was injected in to UFLC system for the analysis. The peak profile and peak purity of both enantiomers are shown in Fig. 5, 6, 7 and 8. Figure 5: Peak Profile Enantiomer 1 Figure 6: Peak Profile Enantiomer 2 Figure 7: Peak Purity Enantiomer 1 Figure 8: Peak Purity Enantiomer 2 RESULTS AND DISCUSSION Validation of the method The developed method for the assay of venlafaxine has been validated as per the current ICH Q2 (R1) guidelines.[11] Analytical parameters The development of NP-UFLC method for the determination of enantiomers has received a considerable attention in recent past because of its importance in the quality control of drugs and drug products. The assay of venlafaxine hydrochloride enantiomers was resolved with good accuracy. The retention time (tR) of R- venlafaxine hydrochloride and S- venlafaxine hydrochloride were found to be 4.5 ±0.2 min and 5.3 ±0.3 min, respectively. A typical chromatogram of R-Venlafaxine hydrochloride and S- venlafaxine hydrochloride is shown in Figure 3. Tailing factor for both R-venlafaxine hydrochloride and S-venlafaxine hydrochloride was found to be 1.1 and 0.8 respectively. The calibration curve was constructed by plotting the peak areas against the concentration of R-and S-venlafaxine hydrochloride in 5-30  µg mL-1 were shown in the Figure 4. It was found to be linear with a correlation coefficient of 0.9971 for R-venlafaxine hydrochloride and 0.9992 for S-venlafaxine hydrochloride, the r epresentative linear regression equation being y = 10507X +2467.1 and y = 10654X +2065.8 for both the enantiomers respectively. The slope, y-intercept, and their standard deviations evaluated are presented in Table 1. Table 1: Regression and sensitivity parameters of enantiomer-1 and enantiomer-2 Accuracy and precision The amount of venlafaxine hydrochloride enantiomers in the matrix was calculated using following formula. % Recovery = T-A /SÃâ€"100 T–total amount of drug estimated, A-initial amount of drug in the tablet powder and S- amount of pure drug added. The results revealed (Table 2), high recovery of Venlafaxine hydrochloride enantiomers, indicating that the proposed method for the determination of venlafaxine hydrochloride enantiomers in the tablet is highly accurate. The intraday and inter day percentage relative standard deviation values were shown in Table 3. These values were within the standard limits. Table 2: Accuracy data of enantiomer-1 and enantiomer-2 Mean value of six determinations Table 3: Precision data of enantiomer-1 and enantiomer-2 Limit of detection and limit of quantification Limit of detection can be calculated using the following equation according to ICH guidelines: LOD = 3.3 x N/S LOQ = 10 x N/S where N is the standard deviation of peak areas of the drug and S is the slope of the corresponding calibration curve. The results are shown in Table 1. Assay of the drug The chiral NP-HPLC method developed in the present investigation was used to quantify venlafaxine hydrochloride enantiomers in tablet dosage forms. The obtained results are given in Tables 4. The average drug content was found to be 10.047 mg for R-venlafaxine hydrochloride and 9.978 mg for S-venlafaxine hydrochloride of the labelled amount in 25mg of racemic venlafaxine hydrochloride, respectively. Table 4: Assay of Venlafaxine Robustness of the method and stability of the solution The robustness of an analytical procedure has been defined by the ICH as a â€Å"measure of its capacity to remain unaffected by small, but deliberate variations in method parameters. The most important aspect of robustness is to develop methods that develop methods that allow for expected variations in method parameters. According to ICH guidelines, robustness should be considered early in the development stage of a method. The typical variations studied under this parameter are flow rate, wavelength and mobile phase composition. The results are tabulated in Table 5. Table 5: Robustness data of enantiomer-1 and enantiomer-2 CONCLUSION A simple, rapid and normal phase chiral UFLC method has been developed and validated for the enantiomeric separation of venlafaxine in tablet formulation. This method is precise, accurate, robust, and specific. Satisfactory results were obtained from the validation of the method. The short retention time (4.5 min for enantiomer 1 and 5.3 for enantiomer 2) obtained provides rapid determination of venlafaxine, which is significant for its routine analysis in quality control. The method exhibits an excellent performance in terms of sensitivity and robust. The experimental results of the present study showed that the proposed NP-UFLC method is simple, specific, precise, sensitive, rapid and accurate and is useful for separation of venlafaxine hydrochloride enantiomers in its pharmaceutical formulation. Death Comes for the Archbishop :: Death Comes for the Archbishop

Death Comes for the Archbishop: A Narrative or a Novel? Is it a narrative or a novel? Few questions have caused me this much trouble. It became apparent to me that Willa Cather's Death Comes for the Archbishop would not be an easy piece of literature to classify. Within just a few pages, it became apparent that this piece of literature is more than either category will allow. While reading the pages of this story I get the feeling that I am not reading a novel, rather it is a series of short stories. I understand why the critics claim that this is not a novel. While the chapters in each section seem to be connected, the sections themselves tend to be disconnected from each other. Each one is able to stand on its own. It is not until I step back and reflect upon the novel that I can see the ties that bind the different sections together. This is not a story about Archbishop Jean Marie Latour, in the end I still do not know him. Rather, Willa Cather uses him as a vessel to display the world around him. It is through him that I learn about the people and places of New Mexico. It is through his eyes the beautiful landscape of the desert comes to life. I can see and smell the land in which he travels. I hear through his ears the legends that make the history of the land so important. It is apparent to me that this is not a novel about an individual person, but about a place. The land and people of New Mexico are the storytellers. It is through them that this series of short stories are joined together into a novel. It is the story of these events that make up the narrative. There is no way to simply classify Death Comes for the Archbishop. It is a novel, a narrative, and a series of short stories. It is simply, what you make of it. Death Comes to the Archbishop could be asserted as something other than a novel. I believe that the stereotypical novel is more structured and rigid in refinement than Cather's example. I see that a strong plot cannot be identified and that the journal approach seems to be misleading. There really is not an antagonist, so in counterpart, there is no protagonist.

Dash Diet Evaluation

Audrey Conn Special Diet Project 04/19/12 The Dash Diet 1. The diet I chose to follow is called the DASH diet. It stands for dietary approaches to stop hypertension (The Dash Diet Eating Plan, 2012). 2. The diet was created for people with hypertension. I followed the diet for three days. Normally I don’t think I would have had a problem following this diet because I tend to eat the same foods already but situational factors affected my outcome. The combination of work and finals didn’t give me adequate time to prepare meals.I tried my hardest to get the fruit, grains, and nuts/seeds/legumes servings in because those were the easiest to eat on the go. I followed that pretty well but I definitely didn’t get all of my vegetable, lean meat/poultry/fish, or dairy servings in. 3. The general purpose of this diet is to prevent and control hypertension, also known as high blood pressure. Besides lowering blood pressure this diet aims to improve insulin sensitivity, lowe r cholesterol, reduce the risk of; cancer, stroke, heart disease, kidney stones, and diabetes, and can also be used as a weight loss diet (The Dash Diet Eating Plan, 2012). . There are a few factors that could potentially make this diet difficult to follow. This diet calls for a lot of whole foods like fruit and vegetables and lean meat. Since this diet is specifically for hypertensive individuals processed foods are out of the question because they contain a large amount of sodium. This detail could pose a problem to those with a hectic lifestyle. Many people don’t have the time to cook or prepare fresh foods. And others simply don’t enjoy it. If a person isn’t willing to avoid the convenience of processed foods than they cannot follow this diet.This also poses a problem for those that are in the lower class. Fresh fruits and vegetables are more expensive than processed foods and some may not be able to afford it. Availability of the fruits and vegetables could be a problem as well. I didn’t have a problem finding them but someone that doesn’t live near a grocery might, or in an area that does not import out of season produce. Your education on nutrition might hinder your success with this diet. You need to know how to read nutrition labels, your calorie/nutritional needs, serving sizes for different foods, and food reparation. Luckily you can buy The DASH Diet Action Plan book on the website that has a lot of helpful information, advice, and recipes. 5. The first difference is that the Dash Diet categorizes food into 7 groups opposed to 5 on the food guide pyramid. The diet separates legumes and meat into 2 different groups. The food guide pyramid also took out a fats and sweets category whereas the Dash Diet still includes it. The fruit, vegetable and dairy food groups were all very similar in serving size. Grain consumption was a couple servings higher in the Dash Diet.Over all protein was also a little higher because the diet puts legumes/seeds/nuts into a separate group. 6. The Dash Diet focuses on low sodium consumption, or more precisely, the correct amount of sodium consumption. The typical eating pattern includes more sodium than our body needs. This diet also has a higher fiber content than most people usually consume. 7. This diet plan is made for long term use. It contains all the necessary vitamins and minerals that our body needs. As long as you switch up the variety in your foods you won’t be lacking or consuming excessive nutrients. 8.Trying to fit enough lean meat and dairy into my diet was the biggest change I had to make. On average I probably get only 2 to 4 servings of lean meat/fish/poultry a week. Tuna was the easiest way for me to get the servings in. My lack of extra time would make this diet hard for me to follow well. My schedule is always changing so preparing foods all the time would be a hassle. It is also more expensive to follow this diet than my typical eating ha bits. Money isn’t something I have a lot of. 9. If I had hypertension and was expected to follow this diet I don’t think the transition would be that difficult.However, the transition would be easier if I had a scheduled time every day or every week that gave me time to prepare food and meals in bulk to save time. 10. I think I’ll have a better understanding of what others go through when they have special diets. It can be a large inconvenience depending on your lifestyle. Down East Spud Busters Case Analysis Essay

This will give this big conglomerate grounded to the â€Å"little guy† and hopefully keep the company connected on the local level as it expands. As Deep East Spud Busters expands from Canada and Maine to Idaho, Europe, the Pacific Rim, and then into nationwide sales and distribution, the â€Å"people factor’ will become increasingly important. By the â€Å"people factor,† I am of course, to put it into business terms, talking about Human Resources. Given the immense geographical expanses and logistical challenges that will no doubt be a factor in such a major expansion, Human Resources are going to very important as they open up the new production plant in Idaho, continue innovating at the Maine higher-value processing plant where new, fresh ideas are paramount, penetrating international markets in Europe and the Pacific Rim where consumers have a totally foreign culture and speak a different language, and as they tap into nationwide sales and distribution here across the United States. The reason being is that folks are different wherever you go and to have a successful company these days, management needs to be less rigid and more organic while treating its stakeholders as individuals with different needs and embracing diversity. But maybe you’re saying to yourself right now â€Å"potatoes are potatoes anywhere you go† or something like that. My response would be that sure potatoes are potatoes but people and their cultural situations are unique wherever you go. Laws and regulations are also different wherever one might go. Legislation, the role of trade unions and governments and their agencies, culture and values vary between regions and countries. Therefore the particular institutional structures operating in different national economies and societies are no doubt influential in my opinion, in maintaining dissimilar Human Resources policies in different countries, American states, counties, and cities. People’s behavior and needs will be influenced by the land in which they reside, and Deep East Spuds Busters is going to have to adjust accordingly. If DESB holds the people factor in high regard as they expand into new territories, they will increase their chance of success exponentially. I have personally have had several jobs where the company couldn’t care less about its employees and it’s not only miserable for the workers, but in my opinion it’s got to be bad for business as well. Company’s like Home Depot, Whittier Wood Products, and literally every wood mill in the Eugene/Springfield area treats their employees like crap, doesn’t pay them enough, overworks them, fires them and lays them off like it’s nothing, and most importantly, those businesses don’t take into account their employees individuality at all. There are definitely companies that not only should follow my idea of how Deep East Spud Busters should maintain their Human Resources departments, but also could be successful in adopting a similar organizational structure to their current and future business models. Some types of companies that I think could benefit from copying DESB’s current and future organizational model are of the following: Land owners who have timber on their land, independent gardeners, independent gold miners, independent mushroom growers, local artisans of pottery, wood crafts, and textiles, the list goes on and on. There are many types of businesses that could follow a template starting with a local cooperative, refinement of received products and materials, nationwide distribution and sales involving local salesmen, and then expansion into international markets. I think companies that produce a product easily made by the individual person could follow this model and not so much highly technical and industrial fields that already have their structures well establishes. For example, it would make no sense for Intel’s microchip producers to follow this model but it would make sense for an individual or small business that creates popular art pieces. Broker: A person who assembles and coordinates participants in a network. Standardization: Establishing common routines and procedures that apply uniformly to everyone. Formalization: The presence of rules and regulations governing how people in the organization interact. Coordination by Plan: Interdependent units are required to meet deadlines and objectives that contribute to a common goal. Business Agility and Ways to Improve It: Business agility is the ability of a business to adapt rapidly and cost efficiently in response to changes in the business environment. Business agility can be maintained by maintaining and adapting goods and services to meet customer demands, adjusting to the changes in a business environment and taking advantage of human resources. Agility is a concept that incorporates the ideas of flexibility, balance, adaptability, and coordination under one umbrella. Put in the context of business, agility refers to the ability of an organization to rapidly adapt to market and environmental changes in productive and cost-effective ways. The agile company is an extension of this concept, referring to an organization that utilizes key principles of adaptive systems and complexity science to achieve success. One can say that business agility is the outcome of Organizational Intelligence. Different areas that enterprises can improve their business agility are through strategy, commitment to customers, and their use of technology. Organization Structure Definition: Definition: The typically hierarchal arrangement of lines of authority, communications, rights and duties of an organization. Organizational structure determines how the roles, power and responsibilities are assigned, controlled, and coordinated, and how information flows between the different levels of management. A structure depends on the organization’s objectives and strategy. In a centralized structure, the top layer of management has most of the decision making power and has tight control over departments and divisions. In a decentralized structure, the decision making power is distributed and the departments and divisions may have different degrees of independence. A company such as Proctor ;amp; Gamble that sells multiple products may organize their structure so that groups are divided according to each product and depending on geographical area as well.
